40% See HS Symptom Reduction in Povorcitinib Study

A new treatment called povorcitinib may help reduce symptoms of hidradenitis suppurativa (HS), based on positive results from two large late-stage clinical trials. In both studies, around 40 percent of people taking povorcitinib had a significant reduction in HS symptoms after 12 weeks, with results better than those taking a placebo (inactive treatment).

Each of the trials included about 600 adults with moderate to severe HS. Researchers tested two doses of povorcitinib — 45 milligrams and 75 milligrams, taken daily. They found that both helped more people reach the primary goal of treatment: at least a 50 percent drop in the number of abscesses (painful, pus-filled lumps) and inflammatory nodules (raised bumps beneath the skin) without any new abscesses or tunnels forming. This response was seen in more than 40 percent of those taking povorcitinib, compared to less than 30 percent of those taking a placebo.

Povorcitinib is a Janus kinase 1 (JAK1) inhibitor, taken by mouth once a day. It works by blocking a pathway involved in inflammation. In addition to reducing skin lesions, the drug also led to faster relief of skin pain, fewer flare-ups, and deeper symptom improvements for many people. These benefits were also seen in people who had previously used biologic drugs without enough relief.

Povorcitinib was generally well tolerated in both studies. Researchers did not find any new safety concerns during the 12-week period, and the safety profile matched what had been seen in earlier trials.

A New Oral Option for HS on the Horizon
